- Stagecoach line opened from St. Louis to Franklin.
- March 6: Missouri Compromise approved by the U.S. Congress.
- March 6: Missouri Enabling Act passed and approved by President Madison.
- June 12-July 19: Missouri Constitutional Convention held, to draw up a Consitution for the State of Missouri.
- November 16: Missouri Legislature creates Lillard County, which included all the land which would become Henry County. (The original Lillard County includes all of what is now Lafayette, Johnson, Henry, the north half of St. Clair, Jackson, Cass, and the north 4/5ths of Bates County.)

- Harmony Mission opened in what is now Bates County.
- February 18: U.S. Congress approves admission of Missouri as a state.
- August 10: President James Monroe proclaims Missouri admission as the 24th State.
- September 1: William Becknell starts on first trading trip to Santa Fe.
- December 31: Site of future state capital at Jefferson City selected.

- Federal Land office opened at Lexington.
- Seat of Lillard County moved from Mount Vernon to the new town of Lexington.

- May: Lexington Township organized, in Lillard County. It included all of what became Henry County.

- April 129: Lafayette visits St. Louis.
- June 2: Great and Little Osage and Kansas Indians ceded their claims to all their lands in Missouri, including what is now Henry County.
- June 2: Osage reliquish all claims to lands in Missouri.
- November 7: Shanwee Indians exchange Missouri land claims for lands elsewhere.

- October 1: Permament seat of government for Missouri established at Jefferson City.
